TheMedicago truncatulahypermycorrhizal B9 mutant displays an altered response to phosphate and is more susceptible toAphanomyces euteiches
2014
Inorganic phosphate (Pi) plays a key role in the development of arbuscular mycorrhizal (AM) symbiosis, which is favoured when Pi is limiting in the environment. We have characterized the Medicago truncatula hypermycorrhizal B9 mutant for its response to limiting (P/10) and replete (P2) Pi. On P2, mycorrhization was significantly higher in B9 plants than in wild-type (WT). The B9 mutant displayed hallmarks of Pi-limited plants, including higher levels of anthocyanins and lower concentrations of Pi in shoots than WT plants. Transcriptome analyses of roots of WT and B9 plants cultivated on P2 or on P/10 confirmed the Pi-limited profile of the mutant on P2 and highlighted its altered response t…
Influence of plant traits, soil microbial properties, and abiotic parameters on nitrogen turnover of grassland ecosystems
2016
International audience; Although it is known that multiple interactions among plant functional traits, microbial properties , and abiotic soil parameters influence the nutrient turnover, the relative contribution of each of these groups of variables is poorly understood. We manipulated grassland plant functional composition and soil nitrogen (N) availability in a multisite mesocosm experiment to quantify their relative effects on soil N turnover. Overall, root traits, arbuscular mycorrhizal colonization, denitrification potential, as well as N availability and water availability, best explained the variation in measured ecosystem properties, especially the trade-off between nutrient sequest…
CRISPR-Cas9 screen reveals a MYCN-amplified neuroblastoma dependency on EZH2.
2018
Pharmacologically difficult targets, such as MYC transcription factors, represent a major challenge in cancer therapy. For the childhood cancer neuroblastoma, amplification of the oncogene MYCN is associated with high-risk disease and poor prognosis. Here, we deployed genome-scale CRISPR-Cas9 screening of MYCN-amplified neuroblastoma and found a preferential dependency on genes encoding the polycomb repressive complex 2 (PRC2) components EZH2, EED, and SUZ12. Genetic and pharmacological suppression of EZH2 inhibited neuroblastoma growth in vitro and in vivo. Moreover, compared with neuroblastomas without MYCN amplification, MYCN-amplified neuroblastomas expressed higher levels of EZH2. ChIP…
Acaulospora sieverdingii, an ecologically diverse new fungus in the Glomeromycota, described from lowland temperate Europe and tropical West Africa
2011
International audience; From a survey of arbuscular mycorrhizal (AM) fungi in agro-ecosystems in Central Europe and West Africa, an undescribed species of Acaulospora was recovered and is presented here under the epithet Acaulospora sieverdingii. Spores of A. sieverdingii are 60-80 mu m in diam, hyaline to subhyaline to rarely light yellow and have multiple pitted depressions on the outer spore wall similar to those known for A. alpina, A. cavernata, A. paulinae and A. scrobiculata. The pits in A. sieverdingii are tiny and often irregular and resemble small dots (0.8-1.8 mu m) or lines (0.5-1.2 x 1.8-2.5 mu m). Analyses of the ITS1, 5.8S subunit and ITS2 regions of the rDNA resolved each of…
Lack of association between proliferative verrucous leukoplakia and human papillomavirus infection.
2005
Purpose To analyze proliferative verrucous leukoplakia (PVL) for the presence of human papillomavirus (HPV) in different stages of the disease. Materials and Methods We studied 13 patients with PVL. In 10 patients (76.9%), a lesional biopsy was taken and frozen at −40°C. Four patients were instructed to mouth rinse with sterile sera. The biopsy and rinse samples were analyzed for HPV by PCR. Results We did not detect HPV infection in the PVL tissue or in the oral rinse of any of the 13 patients in any stage of the disease analyzed, neither in oral squamous cell carcinoma nor in the simple hyperkeratosis. Conclusion There was no association between PVL and HPV infection in our patients.
Helminth communities of herons (Aves: Ardeidae) in southern Italy
2016
The helminth communities of nine species of herons from southern Italy were studied and compared. Of 24 taxa found including seven digeneans, seven nematodes, six cestodes and four acanthocephalans, only five taxa were found in more than one heron species, and five of the 21 taxa that could be identified to species level were classified as 'heron specialists'. The total number of helminth species per heron species ranged from 1 in Botaurus stellaris to 9 in Ixobrychus minutus with infection levels generally low. A statistical comparison was carried out for herons with a sample size >. 5. At the infracommunity level, only I. minutus clearly differed from other heron species. Diversity parame…
Visually induced motion sickness and presence in videogames: The role of sound
2012
Visually induced motion sickness (VIMS) is a well-known phenomenon in virtual environments, simulators, and videogames. We conducted an experiment to analyze the role of sound on the severity of VIMS and the feeling of presence in videogames. Thirty-two subjects first watched a pre-recorded sequence of the game “Mirrors Edge” and then played the game actively. Game-play sound was activated for half of the participants. VIMS was measured via the Simulator Sickness Questionnaire and the Fast Motion Sickness Scale, presence was captured using the Presence Questionnaire. Results showed severe VIMS in all participants during the passive video session, whereas active-play revealed only moderate …
Macrofungal diversity and ecology in two Mediterranean forest ecosystems.
2014
The macrofungal species richness and community assemblages in Italian native woodlands of oaks and Carpinus betulus and non-native woodlands of Pinus spp., Cupressus sempervirens and Eucalyptus camaldulensis were examined through the collection of basidiomata and ascomata over 1 year. The sampling in Collestrada (Umbria) and Pizzo Manolfo (Sicily) forests revealed 216 species of macrofungi. The results indicate differences in macromycete richness and diversity patterns between the two sites. The dominant tree species of the two sites were different; thus, the Collestrada forests had higher mycorrhizal species richness, while the Pizzo Manolfo forest had a higher relative number of saprotrop…
Ligand‐Specific αβ and γδ T Cell Responses in Childhood Tuberculosis
2000
The alphabeta and gammadelta T cell responses were analyzed in the peripheral blood of children affected by active tuberculosis (TB) and in healthy children who tested positive (PPD+) or negative (PPD-) for purified protein derivative. PPD+ healthy and diseased children responded equally well to PPD in vitro. In contrast, only 18% of PPD+ TB patients responded to peptide p38G derived from the 38-kDa protein of Mycobacterium tuberculosis. Analysis of the whole gammadelta T cell population and of its Vgamma9/Vdelta2 subset showed similar frequencies in PPD+ children with TB and in healthy PPD+ and PPD- children. Vgamma9/Vdelta2 cells from children with TB responded to 5 different phosphoantig…
